【导读】2020年7月2日,冷泉港实验室Anthony Zador 等研究组在Cell杂志上发表文章BRICseq bridges brain-wide interregional connectivity to neural activity and gene expression in single animals,报道了BRICseq(Brain-wide individual-animal connectome sequencing)技术,利用DNA条形码(DNA barcodes)和高通量测序技术来解决全脑尺度长距离连接组绘制问题。BRICseq技术基于Zador实验室之前报道的MAPseq (Multiplexed analysis of projections by sequencing)技术【13-15】,并在其基础上进行了改进且极大地扩增了其应用尺度。该技术的关键在于使用大量独特且多样的核酸条形码来标记并追踪单个神经元的长距离投射。相比于传统技术中可被使用并准确检测的颜色数目的局限性,核酸分子不仅仅具有理论上无穷的多样性,并且可以被高通量、高可靠性的测序技术而检测,从而极大的提高了长距离连接组的测绘通量。
